4D Nucleome Project Maps Human Genome’s Three-Dimensional Architecture in Landmark Studies

The National Institutes of Health’s 4D Nucleome Program has published a landmark collection of studies in Science and Science Advances this week, providing the most detailed map yet of how the human genome organizes itself in three-dimensional space and how that architecture changes over time. The research, involving more than three dozen laboratories across eight countries, identifies over 140,000 looping interactions between genes and distant regulatory elements.
The cover story in Science describes how the genome’s folded structure — its “fourth dimension” of nuclear organization — shapes cell identity, shifts across development and aging, and goes awry in disease. The consortium integrated data from more than a dozen experimental techniques including Hi-C, Micro-C, SPRITE, and GAM.
“This is like going from a flat map of the world to a globe showing how continents are actually connected beneath the surface,” said one researcher. “We’ve known that DNA looping matters for gene regulation, but now we can see the full wiring diagram.”
